Effects of Growth on Dinitrogen on the Transcriptome and Predicted Proteome of Nostoc PCC 7120

چکیده
Upon growth on dinitrogen, the filamentous cyanobacterium Nostoc PCC 7120 initiates metabolic and morphological changes. We analyzed the expression of 1249 genes from major metabolic categories under nitrogen fixing and non-nitrogen fixing growth. The expression data were correlated with potential target secondary structures, probe GC-content, predicted operon structures, and nitrogen content of gene products. Of the selected genes, 494 show a more than 2-fold difference in the two conditions analyzed. Under nitrogen-fixing conditions 465 genes, mainly involved in energy metabolism, photosynthesis, respiration and nitrogenfixation, were found to be stronger expressed, whereas 29 genes showed a stronger expression under non-nitrogen fixing conditions. Analysis of the nitrogen content of regulated genes shows that Nostoc PCC 7120 growing on dinitrogen is freed from any constraints to save nitrogen. For the first time the expression of high light-induced stress proteins (HLIP-family) is shown to be linked to the nitrogen availability.

This study focuses on Ultra Violet stress (UVS) gene product which is a UV stress induced protein from cyanobacteria, Synechocystis PCC 6803. Three dimensional structural modeling of target UVS protein was carried out by homology modeling method. 3F2I pdb from Nostoc sp. PCC 7120 was selected as a suitable template protein structure. متن کاملBMAA Inhibits Nitrogen Fixation in the Cyanobacterium Nostoc sp. PCC 7120
Cyanobacteria produce a range of secondary metabolites, one being the neurotoxic non-protein amino acid β-N-methylamino-L-alanine (BMAA), proposed to be a causative agent of human neurodegeneration. As for most cyanotoxins, the function of BMAA in cyanobacteria is unknown. Here, we examined the effects of BMAA on the physiology of the filamentous nitrogen-fixing cyanobacterium Nostoc sp. PCC 71...

The changes in the expression of sigma factor genes during dehydration in terrestrial Nostoc HK-01 and aquatic Anabaena PCC 7120 were determined. The expression of the sigJ gene in terrestrial Nostoc HK-01, which is homologous to sigJ (alr0277) in aquatic Anabaena PCC 7120, was significantly induced in the mid-stage of dehydration. We constructed a higher-expressing transformant of the sigJ gen...
متن کاملHydrogenases and hydrogen metabolism of cyanobacteria.
Cyanobacteria may possess several enzymes that are directly involved in dihydrogen metabolism: nitrogenase(s) catalyzing the production of hydrogen concomitantly with the reduction of dinitrogen to ammonia, an uptake hydrogenase (encoded by hupSL) catalyzing the consumption of hydrogen produced by the nitrogenase, and a bidirectional hydrogenase (encoded by hoxFUYH) which has the capacity to bo...
